I could just be grasping at straws here, but out of any kind of rune and oath combination required if there even is a requirement , everything seems to point me to Beast, Eye, Guidance, and Milkweed the whole brain appearance. There are three Deep Sea runes; the first can be found in the Lower Hintertomb Root Chalice, the second in the Forbidden Woods by two viper pits, and the third can be found in the Isz Root Chalice.
